Virus serotype identified by RT-PCR using type specific primers  and sequencing (Seg-2)   [1].  Confirmed by SNT at NVSL.

(82% similar (Seg-2) to other BTV-3 isolates from Florida

(1130 bp region))

BTV-3

 

(Seg-2 RT-PCR and sequencing, suggests BTV-3, however only 1 set of BTV-3 specific primers out of 5 gave +ve amplification.  82% sequence homology was detected in 675bp of Seg-2 to FL isolates of BTV-3 (which show 91-98% homology to each other).  This places the virus as BTV-3 (Maan et al 2007)

 

A virus neutralisation assay also indicated

 BTV-3

although the type specificity of the BTV-3 sera is unconfirmed.

Original isolate was submitted from the Southeastern Cooperative Wildlife Disease Study (SCWDS) Athens GA. isolate obtained from tissues samples collected in the fall of 2006 in Yalobusha County MS. At SCWDS, samples were innoculated into cattle pulmonary artery endothelial (CPAE) cells cultures. sample sent to NVSL for typing.
